cyclin D1
Summary
Cyclin D1 is a key regulatory protein that controls the G1 to S phase transition of the cell cycle by activating cyclin-dependent kinases 4 and 6 (CDK4/6). It is overexpressed in several malignancies, most notably mantle cell lymphoma, due to the t(11;14) translocation involving the CCND1 gene and IGH locus.
Detail
Cyclin D1 (encoded by CCND1) partners with CDK4/6 to phosphorylate the retinoblastoma protein (Rb). Phosphorylated Rb releases the E2F transcription factor, which then promotes transcription of genes required for S phase entry (e.g., DNA polymerase, thymidine kinase), driving the cell past the G1/S restriction checkpoint. Cyclin D1 expression is normally induced by growth factor signaling (e.g., via the Ras-MAPK pathway) and is a critical link between extracellular mitogenic signals and the cell cycle machinery.
Clinical significance: The t(11;14)(q13;q32) translocation juxtaposes CCND1 with the immunoglobulin heavy chain (IGH) enhancer, leading to constitutive overexpression of cyclin D1. This is the hallmark genetic lesion of mantle cell lymphoma (MCL) and is used diagnostically via immunohistochemistry (positive nuclear staining for cyclin D1) and FISH. Cyclin D1 overexpression is also seen in other cancers including breast cancer, parathyroid adenomas, and some squamous cell carcinomas, contributing to uncontrolled proliferation by bypassing normal cell cycle checkpoints.
Therapeutic relevance: CDK4/6 inhibitors (e.g., palbociclib, ribociclib, abemaciclib) target the cyclin D1-CDK4/6-Rb axis and are used in hormone receptor-positive, HER2-negative breast cancer. Understanding this pathway is essential for grasping oncogenesis, tumor suppressor gene function (Rb), and modern targeted cancer therapies.
Key associations for boards: Cyclin D1 + t(11;14) + CD5+ CD23- B cells = Mantle cell lymphoma. Contrast with cyclin E (G1/S boundary) and other cyclins (A, B) that regulate later phases of the cell cycle.
