zona pellucida
Summary
The zona pellucida is a glycoprotein shell surrounding the oocyte (and early embryo) that mediates species-specific sperm binding, triggers the acrosome reaction, and blocks polyspermy after fertilization (zona reaction). It is shed ("hatching") at the blastocyst stage before implantation.
Detail
STRUCTURE: Extracellular matrix of glycoproteins ZP1, ZP2, ZP3 (and ZP4 in humans), secreted by the oocyte and follicular (granulosa) cells beginning at the primary follicle stage. It lies between the oocyte plasma membrane and the corona radiata (granulosa cells that surround the oocyte).
FERTILIZATION SEQUENCE (high-yield): 1) Capacitated sperm penetrates the corona radiata (hyaluronidase). 2) Sperm binds ZP3 (primary sperm receptor; species-specific). 3) Binding triggers the acrosome reaction: release of acrosin and other proteases that digest a path through the zona. 4) Sperm fuses with the oocyte membrane (Izumo on sperm and Juno on oocyte). 5) Cortical reaction: Ca2+ wave causes cortical granules to release enzymes that cleave/modify ZP2 and ZP3 (zona reaction). This makes the zona impenetrable to other sperm, the slow block to polyspermy. The fast block is membrane depolarization. 6) The oocyte completes meiosis II, and pronuclei form and fuse (syngamy).
DEVELOPMENT: The zona keeps blastomeres together during cleavage and prevents premature implantation, for example in the fallopian tube, which would risk ectopic pregnancy. At the blastocyst stage (about day 5-6), the embryo "hatches" from the zona, enabling implantation (about days 6-10) in the endometrium via the trophoblast.
CLINICAL CORRELATES: Failure of the zona reaction leads to polyspermy, which causes triploidy (e.g., 69,XXY), a nonviable partial mole or early miscarriage. Anti-zona antibodies are a proposed cause of immune infertility. Intracytoplasmic sperm injection (ICSI) bypasses the zona in severe male-factor infertility. Assisted hatching is used in IVF for a thickened zona. Defects in ZP genes can cause empty follicle syndrome or infertility. Zona proteins are studied as targets for immunocontraception.
HIGH-YIELD MNEMONIC/PEARL: ZP3 = sperm binding and acrosome reaction trigger. ZP2 = secondary binding, cleaved after fertilization. Cortical granules = slow block to polyspermy. Day 1 = fertilization in the ampulla; zona is lost around day 5-6 at the blastocyst stage.
